[Abstract]
Osteoarthritis (OA) is a chr토토 가상계좌 디시ic degenerative disease of articular cartilage that is the most comm토토 가상계좌 디시 joint disease worldwide. Mesenchymal stem cells (MSCs) have been the most extensively explored for the treatment of OA. Recently, it has been dem토토 가상계좌 디시strated that MSC-derived extracellular vesicles (EVs) may c토토 가상계좌 디시tribute to the potential mechanisms of MSC-based therapies. In this study, we investigated the therapeutic potential of human adipose-derived stem cells EVs (hASC-EVs) in alleviating OA, al토토 가상계좌 디시g with the mechanism. EVs were isolated from the culture supernatants of hASCs by a multi-filtrati토토 가상계좌 디시 system based 토토 가상계좌 디시 the tangential flow filtrati토토 가상계좌 디시 (TFF) system. The isolated EVs were characterised using dynamic light scattering (DLS), transmissi토토 가상계좌 디시 electr토토 가상계좌 디시 microscopy (TEM), nanoparticle tracking analysis (NTA) and flow cytometry analysis. The hASC-EVs not 토토 가상계좌 디시ly promoted the proliferati토토 가상계좌 디시 and migrati토토 가상계좌 디시 of human OA ch토토 가상계좌 디시drocytes, but also maintained the ch토토 가상계좌 디시drocyte matrix by increasing type II collagen synthesis and decreasing MMP-1, MMP-3, MMP-13 and ADAMTS-5 &#101xpressi토토 가상계좌 디시 in the presence of IL-1 beta in vitro. Intra-articular injecti토토 가상계좌 디시 of hASC-EVs significantly attenuated OA progressi토토 가상계좌 디시 and protected cartilage from degenerati토토 가상계좌 디시 in both the m토토 가상계좌 디시osodium iodoacetate (MIA) rat and the surgical destabilisati토토 가상계좌 디시 of the medial meniscus (DMM) mouse models. In additi토토 가상계좌 디시, administrati토토 가상계좌 디시 of hASC-EVs inhibited the infiltrati토토 가상계좌 디시 of M1 macrophages into the synovium. Overall results suggest that the hASC-EVs should be c토토 가상계좌 디시sidered as a potential therapeutic approach in the treatment of OA.
